convert /fs:ntfs c:

You'll have a more robust filesystem with actual security. How long it takes depends entirely on you - your disk, your CPU, the fullness of the existing FAT32 filesystem.

To accelerate this process as much as possible, take the opportunity to clean off all the crap you've been intending to delete for ages, empty your Temp Internet Files (or Mozilla's cache if you're using the real web browser) and defrag the disk. All of which is Good For You anyway. Then, convert the volume.
